Pediatric anaplastic large cell lymphoma (pALCL) is the most common subtype of pediatric mature T-cell lymphoma and is characterized by the abnormal growth of ALK-positive or ALK-negative T-cells. ALK-positive pALCL is more common and has a worse prognosis.1 Two of the common ALK rearrangements include NPM1::ALK from t(2;5)(p23;q35) and TPM3::ALK from t(1;2)(q25;p23).2 The oligomerization domain from NPM1 or TPM3 is fused to the kinase domain of ALK, leading to ligand-independent ALK activation.3-11 ALK+ ALCL respond well to next-generation ALK inhibitors such as Lorlatinib.12
